Boster Bio offers in-cell western assay service for quantification and visualization of membrane and intracellular proteins.
Boster Bio offers In-Cell Western Blot services – an immunofluorescence-based technology for quantification and visualization of proteins and biomarkers of interest within a specific cell type. In-cell western assays (ICW) measure the infrared fluorescence of a fluorophore-conjugated secondary antibody directed against a primary antibody.
The greatest advantage is that this method is performed in a 96-well plate format, allowing for many replicates and more conditions per experiment, which generates higher quality data. ICW blotting is a convenient method for evaluating cell treatments, while avoiding steps that commonly cause errors in conventional western blots, such as gel loading and membrane transfers during electrophoresis, and lysate prep.
Contact us to coordinate the shipment of your cells for assay the day of arrival. Please seed, treat, and fix cells prior to shipping overnight on dry ice. We use Greiner Bio-One plates: #6545097, (Clear bottom tissue culture plates with black walls). Please use ice cold methanol to fix the cells at -20C for 20 minutes.
Boster will perform the assay. We stain the cells with a DNA stain to demonstrate that there are sufficient cells to be evaluated. If you want at least one reliable antibody staining we can use GAPDH to demonstrate that the method works. We can also procure the antibodies for you.
We can detect up to two targets plus total cell normalization using spectrally distinct fluorescent conjugates.
