Background of Smad2
Smad2 (also known as Mothers against decapentaplegic homolog 2, Mothers against DPP homolog 2, Mad2, hMAD-2 or hSMAD2) is a member of the Smad family of proteins which are similar to the gene products of the Drosophila gene 'mothers against decapentaplegic' (Mad) and the C. elegans gene Sma. SMAD proteins are signal transducers and transcriptional modulators that mediate multiple signaling pathways. This protein mediates the signal of the transforming growth factor (TGF)-beta, and thus regulates multiple cellular processes, such as cell proliferation, apoptosis, and differentiation. This protein is recruited to the TGF-beta receptors through its interaction with the SMAD anchor for receptor activation (SARA) protein. In response to TGF-beta signal, this protein is phosphorylated by the TGF-beta receptors. The phosphorylation induces the dissociation of this protein with SARA and the association with the family member SMAD4. The association with SMAD4 is important for the translocation of this protein into the nucleus, where it binds to target promoters and forms a transcription repressor complex with other cofactors. This protein can also be phosphorylated by activin type 1 receptor kinase, and mediates the signal from the activin. Alternatively, spliced transcript variants have been observed for this gene. SMAD2 may act as a tumor suppressor in colorectal carcinoma. It positively regulates PDPK1 kinase activity by stimulating its dissociation from the 14-3-3 protein YWHAQ which acts as a negative regulator. SMAD2 may be associated with diseases such as Keloids and Ureteral Disease. Anti-SMAD2 Antibody is useful for researchers interested in SMAD pathways, transcription factor activity, sequence-specific DNA binding, and cancer research.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-SMAD2 antigen affinity purified polyclonal antibody (Catalog # A00090) at 0.5 ug/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-Rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # SA1022) with Tanon 5200 system. A specific band was detected for SMAD2.

From what I have seen in literature cervix carcinoma erythroleukemia does express SMAD2. From what I have seen in Uniprot.org, SMAD2 is expressed in kidney, placenta, kidney, pancreas spleen, chronic myeloid leukemia cell, colon adenocarcinoma, cervix carcinoma, cervix carcinoma erythroleukemia, among other tissues. Regarding which tissues have SMAD2 expression, here are a few articles citing expression in various tissues:

It shows on the product datasheet, A00090 anti-SMAD2 antibody as been tested on IP. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

We do not advise storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free A00090 anti-SMAD2 antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
