Background of NFATC1/NFAT2

NFATC1 (Nuclear factor of activated T-cells, cytoplasmic 1), also known as NFATC or NFAT2, is a protein that in humans is encoded by the NFATC1 gene. And it is also a component of the nuclear factor of activated T cells DNA-binding transcription complex. The NFATC1 gene is mapped on 18q23. Proteins belonging to this family of transcription factors play a central role in inducible gene transcription during immune response. NFATC1 was not detected in brain, liver, or kidney. NFATC1 is an inducible nuclear component which functions as a major molecular target for the immunosuppressive drugs such as cyclosporin A. The solution structure of the binary complex formed between the core DNA-binding domain of human NFATC1 and the 12-bp oligonucleotide duplex containing the ARRE2 DNA site from the IL2 promoter. Nfatc1 was expressed exclusively in mouse hair follicle stem cells, and using gain- and loss-of-function approaches, and Nfatc1 inhibited stem cell activation.

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

4 Customer Q&As for Anti-NFAT2/NFATC1 Antibody Picoband®

Question

We are currently using anti-NFAT2/NFATC1 antibody A00340-2 for human tissue, and we are satisfied with the ELISA results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on monkey tissues as well?

Asked: 2019-07-24

Answer

The anti-NFAT2/NFATC1 antibody (A00340-2) has not been tested for cross reactivity specifically with monkey tissues, but there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in monkey you can get your next antibody for free. Please contact me if I can help you with anything.

Question

My boss were satisfied with the WB result of your anti-NFAT2/NFATC1 antibody. However we have observed positive staining in peripheral blood lymphocyte t-cell cytoplasm. using this antibody. Is that expected? Could you tell me where is NFATC1 supposed to be expressed?

K. Rodriguez

Asked: 2018-05-30

Answer

From what I have seen in literature, peripheral blood lymphocyte t-cell does express NFATC1. Generally NFATC1 expresses in cytoplasm. Regarding which tissues have NFATC1 expression, here are a few articles citing expression in various tissues:
B-cell, Pubmed ID: 8702849
B-cell lymphoma, Pubmed ID: 10072078
Brain, Pubmed ID: 15489334

Question

We have tried in the past anti-NFAT2/NFATC1 antibody for ELISA on muscle organ in a previous project. I am using mouse, and I plan to use the antibody for WB next. I am interested in examining muscle organ as well as peripheral blood lymphocyte t-cell in our next experiment. Could you please give me some suggestion on which antibody would work the best for WB?

Asked: 2013-05-14

Answer

I looked at the website and datasheets of our anti-NFAT2/NFATC1 antibody and it seems that A00340-2 has been validated on mouse in both ELISA and WB. Thus A00340-2 should work for your application. Our Boster satisfaction guarantee will cover this product for WB in mouse even if the specific tissue type has not been validated. We do have a comprehensive range of products for WB detection and you can check out our website bosterbio.com to find out more information about them.
