Background of NFATC1/NFAT2
NFATC1 (Nuclear factor of activated T-cells, cytoplasmic 1), also known as NFATC or NFAT2, is a protein that in humans is encoded by the NFATC1 gene. And it is also a component of the nuclear factor of activated T cells DNA-binding transcription complex. The NFATC1 gene is mapped on 18q23. Proteins belonging to this family of transcription factors play a central role in inducible gene transcription during immune response. NFATC1 was not detected in brain, liver, or kidney. NFATC1 is an inducible nuclear component which functions as a major molecular target for the immunosuppressive drugs such as cyclosporin A. The solution structure of the binary complex formed between the core DNA-binding domain of human NFATC1 and the 12-bp oligonucleotide duplex containing the ARRE2 DNA site from the IL2 promoter. Nfatc1 was expressed exclusively in mouse hair follicle stem cells, and using gain- and loss-of-function approaches, and Nfatc1 inhibited stem cell activation.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: mouse thymus tissue lysates,
Lane 2: human 22RV1 whole cell lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-NFAT2 antigen affinity purified polyclonal antibody (Catalog # A00340-1) at 0.5 ug/mL overnight at 4 then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for NFAT2 at approximately 101KD. The expected band size for NFAT2 is at 101KD.

Our lab used your anti-NFAT2/NFATC1 antibody for Flow Cytometry on b-cell last year. I am using rat, and We intend to use the antibody for WB next. I am interested in examining b-cell as well as peripheral blood lymphocyte t-cell in our next experiment. Could you please give me some suggestion on which antibody would work the best for WB?

I have checked the website and datasheets of our anti-NFAT2/NFATC1 antibody and I see that A00340-1 has been tested on rat in both Flow Cytometry and WB. Question
My boss were happy with the WB result of your anti-NFAT2/NFATC1 antibody. However we have observed positive staining in b-cell lymphoma cytoplasm. using this antibody. Is that expected? Could you tell me where is NFATC1 supposed to be expressed?

Based on literature, b-cell lymphoma does express NFATC1. Generally NFATC1 expresses in cytoplasm. Regarding which tissues have NFATC1 expression, here are a few articles citing expression in various tissues:

We are currently using anti-NFAT2/NFATC1 antibody A00340-1 for mouse tissue, and we are content with the ELISA results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on monkey tissues as well?

The anti-NFAT2/NFATC1 antibody (A00340-1) has not been validated for cross reactivity specifically with monkey tissues, though there is a good chance of cross reactivity. Based on literature b-cell lymphoma does express NFATC1. Based on Uniprot.org, NFATC1 is expressed in muscle organ, peripheral blood lymphocyte t-cell, b-cell, b-cell lymphoma, brain, erythroleukemia, among other tissues.
