Background of uPAR

The urokinase-type plasminogen activator receptor (uPAR) is a key molecule in the regulation of cell-surface plasminogen activation and, as such, plays an important role in many normal as well as pathological processes. The cDNA for Mo3, an activation antigen expressed by human monocytes and myelomonocytic cell lines after stimulation by a variety of agents. Mo3 expression in vivo is associated predominantly with macrophages in inflammatory sites. It is a highly glycosylated protein of about 50 kD in monocytes where it is anchored to the plasma membrane by glycosyl-phosphatidylinositol linkage. The complete coding sequence of the cDNA has been found to encode 335 amino acids including a predicted signal peptide of 22 residues and a hydrophobic C-terminal portion. Mo3 is identical to the human receptor for the urokinase plasminogen activator. UPAR is a useful prognostic marker for biologically aggressive forms of endometrial cancer. PLAUR is located at chromosome 19q13.1-q13.2. The standard product used in this kit is recombinant human uPAR, consisting of 287 amino acids with the molecular mass of 31KDa.

Intra Assay Consistency & Inter Assay Consistency

We measured random samples of Human uPAR / URKR / uPA Receptor ELISA Kit PicoKine® within the same batch/lot to ensure the consistency of the kits' performances. ELISA intra assay consistency is measured using wells from the same plate/assay kit. ELISA inter assay consistency is measured using wells from different plates from the same batch production/lot.

5 Customer Q&As for Human uPAR / URKR / uPA Receptor ELISA Kit PicoKine®

Question

Q: how long can samples (cell cultures, serum, and plasma) be stored and still be stable for quantification uPAR using the EK0536 Human uPAR Picokine® ELISA Kit?

Verified Customer

Verified customer

Asked: 2020-02-20

Answer

A: Boster Bio does not assess sample stability. Variations in sample collection, processing, and storage may affect the stabilityof samples. It is recommend to assay sample on priority basis collection when possible, or aliquot into single use volumes and store samples frozen. limit repetitive freeze-thaw cycles with the stored samples to limit protein degradation.

Question

Q: is it okay to use citrate plasma as samples in Human uPAR Picokine® ELISA Kit (Catalog # EK0536)?

Verified Customer

Verified customer

Asked: 2018-05-12

Answer

A: Chelating agents such as EDTA, Heparin and Citrate can attach metal ions from the functional domain of uPAR causing disruption of its protein structure. uPAR may be denatured as a result and may compromise the assay's measurements. The chilating sites could also be too close to the epitopes a must for detection and block the antigen antibody reaction. We have tested the uPAR ELISA, treating samples with different anticoagulants and decided that heparin or EDTA can be used for treatment of blood/plasma samples. Do not use other anticoagulents when collecting samples.

Question

Q: Are Boster Bio recombinant proteins and antibodies sterile?

M. Mehta

Verified customer

Asked: 2014-10-19

Answer

A: although the vials are bottled using aseptic techniques, heat-treated vials, and sterile stock solutions, they are not considered or guaranteed to be sterile. If sterile material is required for an experiment, the material can be filtered through a 0.2 micron filter designed for use with biological fluids.
