The Estrogen Receptor-alpha (Phospho-Ser167) Cell-Based ELISA Kit is a convenient, lysate-free, high throughput and sensitive assay kit that can monitor Estrogen Receptor-alpha (Phospho-Ser167) protein expression profile in cells. The kit can be used for measuring the relative amounts of Estrogen Receptor-alpha (Phospho-Ser167) in cultured cells as well as screening for the effects that various treatments, inhibitors (ie. siRNA or chemicals), or activators have on Estrogen Receptor-alpha (Phospho-Ser167).

- Microplate reader able to measure absorbance at 450 nm and/or 595 nm for Crystal Violet Cell Staining (Optional)
- Micropipettes with capability of measuring volumes ranging from 1 ul to 1 ml
- 37% formaldehyde (Sigma Cat# F-8775) or formaldehyde from other sources
- Deionized or sterile water
- Squirt bottle, manifold dispenser, multichannel pipette reservoir or automated microplate washer
- Graph paper or computer software capable of generating or displaying logarithmic functions

I purchased your product Estrogen Receptor-alpha (Phospho-Ser167) Colorimetric Cell-Based ELISA Kit (EKC2404) and performed Western blot assay but the results shows multiple bands and wrong band size. I need your help to determine the correct size of Western blot image.

Answer
We understand that you expect to see the receptor at 65kDA but you did not. These are the few things we want to ask; what are the samples you used? How did you prepare RIPA lysis buffer? Are they collected in protease inhibitors? Did you boil the sample to denature with the reducing agent? How are the membranes blocked? What is the dilution or concentration of the antibody? What are the sizes (molecular weights) of the bands that you observed? Looking forward your reply to resolve the issue.

We recommend a dilution range in IHC-P is 1:20 to 1:80 and this Estrogen Receptor-alpha (Phospho-Ser167) Colorimetric Cell-Based ELISA Kit requires antigen retrieval with citrate buffer pH 6.0.
