Storage & Handling

Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use. Expiration date is one (1) year from date of opening. (Ship on dry ice.)

Background of SMARCAL1
SmarcAL1 (SWI/SNF-related µatrix-associated actin-dependent regulator of chromatin a-like 1) is also known as HARP, HepA Related Protein, HHARP, SMARCA like Protein 1 and SWI/SNF Related. SmarcAL1 is a member of the SWI/SNF family of proteins. Members of this family have helicase and ATPase activities and are thought to regulate transcription of certain genes by altering the chromatin structure around those genes. The encoded protein shows sequence similarity to the E. coli RNA polymerase-binding protein HepA. Mutations in this gene are a cause of Schimke immunoosseous dysplasia (SIOD), an autosomal recessive disorder with the diagnostic features of spondyloepiphyseal dysplasia, renal dysfunction, and T-cell immunodeficiency.

Western blot analysis of SmarcAL1 expression in HeLa nuclear extract (lane 1), HeLa whole cell lysates (lane 2), A431 whole cell lysates (lane 3), Jurkat whole cell lysates (lane 4) and 293 whole cell lystaes (lane 5). SmarcAL1 at 106KD was detected using rabbit anti-SmarcAL1 Antigen Affinity purified polyclonal antibody (Catalog # A03383) at 1:500. The blot was developed using chemiluminescence (ECL) method (Catalog # EK1002).

Question
My question regarding product A03383, anti-SmarcAL1 antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

We suggest not storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free A03383 anti-SmarcAL1 antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.

Our lab technicians have not tested anti-SmarcAL1 antibody A03383 on pig. You can run a BLAST between pig and the immunogen sequence of anti-SmarcAL1 antibody A03383 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated pig samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in pig lung lymph in WB, you can get your next antibody for free.
