Background of SETDB2
Setdb2 and the neighboring gene Phf11 were linked to familial atopy/ asthma and are in a "critical region" commonly deleted in CLL. Setdb2 is a candidate histone methyltransferase as a member of the Su(var) 3-9 family of SET domain-containing enzymes. Other predicted domains in Setdb2 include a methylcytosine binding domain and a large bifurcation domain interrupting the putative histone methyltransferase (SET) domain whose function is unknown. Anti-Setdb2 antibody is ideal for researchers interested in Cancer Research and Epigenetics Research.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Setdb2 antigen affinity purified polyclonal antibody (Catalog # A08290) at 0.5 ug/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-Rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # SA1022) with Tanon 5200 system. A specific band was detected for Setdb2.

We are currently using anti-Setdb2 antibody A08290 for human tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, mouse. Is it likely that the antibody can work on horse tissues as well?

You can see on the product datasheet, A08290 anti-Setdb2 antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
