Background of MUC6
Mucin 6, oligomeric mucus/gel-forming, also known as MUC6, is a human gene. This gene encodes a member of the mucin protein family. Mucins are high molecular weight glycoproteins produced by many epithelial tissues. The protein encoded by this gene is secreted and forms an insoluble mucous barrier that protects the gut lumen. The MUC6 gene encodes gastric mucin, a secreted glycoprotein that plays an essential role in epithelial cytoprotection from acid, proteases, pathogenic microorganisms, and mechanical trauma in the gastrointestinal tract.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: mouse stomach tissue lysates,
Lane 2: human SGC-7901 whole cell lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Gastric Mucin antigen affinity purified polyclonal antibody (Catalog # A02143) at 0.5 μg/mL overnight at 4℃, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Gastric Mucin at approximately 257KD. The expected band size for Gastric Mucin is at 257KD.

We are currently using anti-Gastric Mucin/MUC6 antibody A02143 for mouse tissue, and we are satisfied with the ELISA results. The species of reactivity given in the datasheet says human, mouse. Is it true that the antibody can work on primate tissues as well?

The anti-Gastric Mucin/MUC6 antibody (A02143) has not been tested for cross reactivity specifically with primate tissues, though there is a good chance of cross reactivity.
