Background of FGF-23
Fibroblast growth factor 23 or FGF23 is a protein that in humans is encoded by the FGF23 gene. This gene encodes a member of the fibroblast growth factor family of proteins, which possess broad mitogenic and cell survival activities and are involved in a variety of biological processes. The product of this gene regulates phosphate homeostasis and transport in the kidney. The full-length, functional protein may be deactivated via cleavage into N-terminal and C-terminal chains. Mutation of this cleavage site causes autosomal dominant hypophosphatemic rickets (ADHR). Mutations in this gene are also associated with hyperphosphatemic familial tumoral calcinosis (HFTC).

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: mouse testis tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-FGF 23 antigen affinity purified polyclonal antibody (Catalog # PB9868) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for FGF 23 at approximately 34 kDa. The expected band size for FGF 23 is at 28 kDa.

We are currently using anti-FGF23 antibody PB9868 for mouse tissue, and we are content with the WB results. The species of reactivity given in the datasheet says mouse. Is it possible that the antibody can work on bovine tissues as well?

The anti-FGF23 antibody (PB9868) has not been validated for cross reactivity specifically with bovine tissues, but there is a good chance of cross reactivity.
