Background of CREB
CREB1, also known as cAMP responsive element binding protein 1, is a protein that in humans is encoded by the CREB1 gene. It is mapped to 2q33.3. This protein binds the cAMP response element, a DNA nucleotide sequence present in many viral and cellular promoters. The binding of CREB1 stimulates transcription. This protein is a CREB transcription factor that is a member of the leucine zipper family of DNA-binding proteins. This protein binds as a homodimer to the cAMP-responsive element, an octameric palindrome. The protein is phosphorylated by several protein kinases, and induces transcription of genes in response to hormonal stimulation of the cAMP pathway.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: recombinant human CREB protein 0.5 ng.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CREB antigen affinity purified polyclonal antibody (Catalog # PB9100) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for CREB at approximately 38 kDa. The expected band size for CREB is at 38 kDa.

From what I have seen in literature testis does express CREB1. From what I have seen in Uniprot.org, CREB1 is expressed in lung, testis, eye, embryonic kidney, cervix carcinoma, leukemic t-cell, cervix carcinoma erythroleukemia, liver, among other tissues. Regarding which tissues have CREB1 expression, here are a few articles citing expression in various tissues:

We are currently using anti-CREB/CREB1 antibody PB9100 for human tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, rat. Is it likely that the antibody can work on bovine tissues as well?
