Background of BARD1
Probable E3 ubiquitin-protein ligase. The BRCA1-BARD1 heterodimer specifically mediates the formation of 'Lys-6'-linked polyubiquitin chains and coordinates a diverse range of cellular pathways such as DNA damage repair, ubiquitination and transcriptional regulation to maintain genomic stability. Plays a central role in the control of the cell cycle in response to DNA damage. Acts by mediating ubiquitin E3 ligase activity that is required for its tumor suppressor function. Also forms a heterodimer with CSTF1/CSTF-50 to modulate mRNA processing and RNAP II stability by inhibiting pre-mRNA 3' cleavage.

Question
Would A01646 anti-BARD1 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Answer
You can see on the product datasheet, A01646 anti-BARD1 antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

We are currently using anti-BARD1 antibody A01646 for human tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says human. Is it possible that the antibody can work on dog tissues as well?

The anti-BARD1 antibody (A01646) has not been validated for cross reactivity specifically with dog tissues, though there is a good chance of cross reactivity.
