A. Immunofluorescence of rat cerebellum section stained with mouse mAb to aldolase C, M05296, dilution 1:1,000, in green, and costained with rabbit pAb to GFAP, dilution 1:5,000 in red. In both A and B blue is Hoechst staining of nuclear DNA. Aldolase C antibody selectively labels the perikarya and dendrites of Purkinje cells, while GFAP antibody stains processes of Bergman glia and astrocytic cells. B. Immunofluorescence of E20 rat cortical culture stained with M05296, dilution 1:1,000 in green, and costained with chicken pAb to MAP2, dilution 1:10,000 in red. The aldolase C antibody labels cytosolic protein expressed in glial cells, while MAP2 antibody stains dendrites and perikarya of mature neurons.

Question
Does M05296 anti-Aldolase C: N-terminus Specific Monoclonal antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
Verified Customer
Verified customer
Asked: 2018-08-22
Answer
As indicated on the product datasheet, M05296 anti-Aldolase C: N-terminus Specific Monoclonal antibody as been validated on IF. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
