Background of PD-L1
Programmed death-ligand 1 (PD-L1), also known as cluster of differentiation 274 (CD274) or B7 homolog 1 (B7-H1), is a protein that in humans is encoded by the CD274 gene. PD-L1 is a 40kDa type 1 transmembrane protein that has been speculated to play a major role in suppressing the immune system during particular events such as pregnancy, tissue allografts, autoimmune disease and other disease states such as hepatitis. Normally the immune system reacts to foreign antigens where there is some accumulation in the lymph nodes or spleen which triggers a proliferation of antigen-specific CD8+ T cell. The formation of PD-1 receptor / PD-L1 or B7.1 receptor /PD-L1 ligand complex transmits an inhibitory signal which reduces the proliferation of these CD8+ T cells at the lymph nodes and supplementary to that PD-1 is also able to control the accumulation of foreign antigen specific T cells in the lymph nodes through apoptosis which is further mediated by a lower regulation of the gene Bcl-2.

A: Boster Bio does not evaluate sample stability. Variations in sample collection, processing, and storage may alter the stabilityof samples. It is recommend to assay sample right after collection when possible, or aliquot into single use volumes and store samples frozen. limit repetitive freeze-thaw cycles with the stored samples in order to limit protein degradation.

Our lab technicians worked on lysates from human PBMCs and conditioned media from unstimulated human PBMCs using the Human PD-L1/B7-H1 ELISA Kit PicoKine™ (EK1449) and Mouse PD-L1/B7-H1 ELISA Kit PicoKine™ (EK1450). The concentration of PD-L1 in human PBMCs lysate is around 600pg/mL-700pg/mL while that of PD-L1 in the culture media is around 80-100pg/mL. Our lab technicians think that EK1449 and EK1450 are able to detect total PD-L1 in samples.

Q: we need your suggestion regarding the dilution ratio of serum samples for detection of PD-L1 in Mouse plasma? I am trying to measure a few analytes and it requires 100ul of diluted samples for each well. We have limited sample volumes so we like to dilute as much as possible.

A: having little idea about the physiological or pathological context of your samples we cannot recommend a dilution ratio without performing a pilot test with your samples. Here is how you can perform a pilot study on your own: perform a serial dilution of your samples on the PD-L1 ELISA kit to make sure you have a linear ascending curve followed by a plateau, which signifies the samples saturating the detection limit of the kit. Then you can pick the dilution ratios from samples in the linear part of the curve as your experimental dilution ratio.

A: do not freeze and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test PD-L1 for a later time, you should let the blood clot in glass tubes and separate the serum to freeze for later analysis.
