Background of SP4
SP4 (Sp4 transcription factor), also known as Transcription factor Sp4, MGC130008, MGC130009, SPR-1, HF1B, is a protein that in humans is encoded by the SP4 gene. It is a multigene family of SP proteins. Northern blot analysis that SP4 is abundantly expressed as a 7.5-kb message in brain, but barely detectable in other human cell lines. SP4 is an activator protein like SP1. However, in contrast to SP1, SP4 is not able to act synergistically through adjacent binding sites.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Rat Brain Tissue Lysate at 50ug,
Lane 2: Mouse Brain Tissue Lysate at 50ug,
Lane 3: U87 Whole Cell Lysate at 40ug,
Lane 4: HELA Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-SP4 antigen affinity purified polyclonal antibody (Catalog # PB9558) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for SP4 at approximately 82 kDa. The expected band size for SP4 is at 82 kDa.

We are currently using anti-Transcription factor Sp4 antibody PB9558 for rat tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on feline tissues as well?
