Background of SP4
SP4 (Sp4 transcription factor), also known as Transcription factor Sp4, MGC130008, MGC130009, SPR-1, HF1B, is a protein that in humans is encoded by the SP4 gene. It is a multigene family of SP proteins. Northern blot analysis that SP4 is abundantly expressed as a 7.5-kb message in brain, but barely detectable in other human cell lines. SP4 is an activator protein like SP1. However, in contrast to SP1, SP4 is not able to act synergistically through adjacent binding sites.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human Jurkat whole cell lysates,
Lane 2: human K562 whole cell lysates,
Lane 3: rat brain tissue lysates,
Lane 4: mouse brain tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Sp4 antigen affinity purified polyclonal antibody (Catalog # PA1865) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Sp4 at approximately 82 kDa. The expected band size for Sp4 is at 82 kDa.

Our lab technicians have not tested anti-Transcription factor Sp4 antibody PA1865 on zebrafish. You can run a BLAST between zebrafish and the immunogen sequence of anti-Transcription factor Sp4 antibody PA1865 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated zebrafish samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in zebrafish uterus in WB, you can get your next antibody for free.
