Background of RPS3
40S ribosomal protein S3 is a protein that in humans is encoded by the RPS3 gene. Ribosomes, the organelles that catalyze protein synthesis, consist of a small 40S subunit and a large 60S subunit. Together these subunits are composed of 4 RNA species and approximately 80 structurally distinct proteins. This gene encodes a ribosomal protein that is a component of the 40S subunit, where it forms part of the domain where translation is initiated. The protein belongs to the S3P family of ribosomal proteins. Studies of the mouse and rat proteins have demonstrated that the protein has an extraribosomal role as an endonuclease involved in the repair of UV-induced DNA damage. The protein appears to be located in both the cytoplasm and nucleus but not in the nucleolus. Higher levels of expression of this gene in colon adenocarcinomas and adenomatous polyps compared to adjacent normal colonic mucosa have been observed. This gene is co-transcribed with the small nucleolar RNA genes U15A and U15B, which are located in its first and fifth introns, respectively. As is typical for genes encoding ribosomal proteins, there are multiple processed pseudogenes of this gene dispersed through the genome. Multiple alternatively spliced transcript variants encoding different isoforms have been found for this gene.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human placenta tissue lysates,
Lane 2: human HL-60 whole cell lysate,
Lane 3: human Caco-2 whole cell lysate,
Lane 4: human K562 whole cell lysate,
Lane 5: human U2OS whole cell lysate,
Lane 6: human MCF-7 whole cell lysate,
Lane 7: human PC-3 whole cell lysate.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-RPS3 antigen affinity purified polyclonal antibody (Catalog # A01542-2) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for RPS3 at approximately 31KD. The expected band size for RPS3 is at 27KD.

The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1μg/ml rabbit anti-RPS3 Antibody (A01542-2) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC)(Catalog # SA1022) with DAB as the chromogen.

We are currently using anti-RPS3 antibody A01542-2 for rat tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on monkey tissues as well?

The anti-RPS3 antibody (A01542-2) has not been validated for cross reactivity specifically with monkey tissues, but there is a good chance of cross reactivity.
