Background of RNF123
E3 ubiquitin-protein ligase RNF123 is an enzyme that in humans is encoded by the RNF123 gene. It is mapped to 3p21.31. The protein encoded by this gene contains a C-terminal RING finger domain, a motif present in a variety of functionally distinct proteins and known to be involved in protein-protein and protein-DNA interactions, and an N-terminal SPRY domain. This protein displays E3 ubiquitin ligase activity toward the cyclin-dependent kinase inhibitor 1B which is also known as p27 or KIP1. Alternative splicing results in multiple transcript variants.

RNF123 was detected in paraffin-embedded section of human intestinal cancer tissues. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1μg/ml rabbit anti-RNF123 Antibody (A09642-1) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC)(Catalog # SA1022) with DAB as the chromogen.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human placenta tissue lysates,
Lane 2: human THP-1 whole cell lysates,
Lane 3: human HepG2 whole cell lysates,
Lane 4: human PC-3 whole cell lysates,
Lane 5: human U-87MG whole cell lysates,
Lane 6: human K562 whole cell lysates,
Lane 7: human Hela whole cell lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-RNF123 antigen affinity purified polyclonal antibody (Catalog # A09642-1) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for RNF123 at approximately 149KD. The expected band size for RNF123 is at 149KD.

We are currently using anti-RNF123 antibody A09642-1 for human tissue, and we are satisfied with the IHC-P results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on goat tissues as well?

Our lab technicians have not validated anti-RNF123 antibody A09642-1 on monkey. You can run a BLAST between monkey and the immunogen sequence of anti-RNF123 antibody A09642-1 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated monkey samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in monkey eye in ELISA, you can get your next antibody for free.
