Background of SERPINB9
Anti-Glycogen Synthase 1 pS641 antibody is validated by IHC, Western Blot and ELISA. Human muscle glycogen synthase (GS) is responsible for the biosynthesis of glycogen from phosphorylated glucose units. Mammalian liver and muscle contain GS consisting of four subunits with a total molecular weight of 360,000. GS is subject to regulation through both allosteric and covalent modification and occurs in two forms: the phosphorylated inactive form, and the dephosphorylated active form. GS is inactivated by the serine/threonine kinase called glycogen synthase kinase-3 that mainly functions to phosphorylate muscle glycogen synthase. This antibody is specific for the phosphorylated form of GS at S641. Phosphorylation of GS at S641 has been associated with Antiphospholipid Antibody Syndrome.

Assay Dilutions Recommendation

Immunohistochemistry (formaldehyde-fixed paraffin-embedded human tissues after pretreatment with 10mM citric acid pH 6.0 for 20min; use ~1:100. For fixed cytospin preparations fix cells for 10min in 10% formalin and then treat them as paraffin sections)

You can see on the product datasheet, M04734 anti-Proteinase inhibitor 9 (human) Monoclonal antibody (PI9-17) as been tested on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

We are currently using anti-Proteinase inhibitor 9 (human) Monoclonal antibody (PI9-17) M04734 for human tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human. Is it possible that the antibody can work on bovine tissues as well?

The anti-Proteinase inhibitor 9 (human) Monoclonal antibody (PI9-17) (M04734) has not been validated for cross reactivity specifically with bovine tissues, but there is a good chance of cross reactivity.
