Background of MMP-10
Stromelysin-2 also known as matrix metalloproteinase-10 (MMP-10) or transin-2 is an enzyme that in humans is encoded by the MMP10 gene. Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. The enzyme encoded by this gene degrades proteoglycans and fibronectin. The gene is part of a cluster of MMP genes which localize to chromosome 11q22.3.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Human Placenta Tissue Lysate at 50ug,
Lane 2: HELA Whole Cell Lysate at 40ug,
Lane 3: SGC Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-MMP10 antigen affinity purified polyclonal antibody (Catalog # PB9670) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for MMP10 at approximately 54 kDa. The expected band size for MMP10 is at 54 kDa.

We are currently using anti-MMP10 antibody PB9670 for human tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human. Is it true that the antibody can work on goat tissues as well?

The anti-MMP10 antibody (PB9670) has not been validated for cross reactivity specifically with goat tissues, though there is a good chance of cross reactivity.
