Background of Periostin/OSF-2
POSTN (Periostin), also known as PN or OSF2, is a protein that in humans is encoded by the POSTN gene. The International Radiation Hybrid Mapping Consortium mapped the POSTN gene to chromosome 13. It was found that purified recombinant PN supported adhesion of ovarian epithelial cells. And periostin was overexpressed by the majority of human primary breast cancers examined. Transfected tumor cell lines overexpressing periostin showed accelerated growth and angiogenesis as xenografts in immunocompromised animals. It was showed that extracellular periostin induced reentry of differentiated mammalian cardiomyocytes into the cell cycle.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: rat gaster tissue lysates,
Lane 2: rat lung tissue lysates,
Lane 3: rat NRK whole cell lysates,
Lane 4: mouse gaster tissue lysates,
Lane 5: mouse intestine tissue lysates,
Lane 6: mouse lung tissue lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Periostin antigen affinity purified polyclonal antibody (Catalog # A01378) at 0.5 ug/mL overnight at 4 then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Periostin at approximately 93KD. The expected band size for Periostin is at 93KD.

We bought anti-Periostin/Postn antibody for ELISA on pubmed:8363580} in a previous experiment. I am using rat, and I plan to use the antibody for IHC next. My question regards examining pubmed:8363580} as well as pubmed:8363580} rc placenta eco:0000269 in our next experiment. Do you have any suggestion on which antibody would work the best for IHC?

I looked at the website and datasheets of our anti-Periostin/Postn antibody and it seems that A01378 has been tested on rat in both ELISA and IHC. Thus A01378 should work for your application. Our lab were satisfied with the WB result of your anti-Periostin/Postn antibody. However we have observed positive staining in osteosarcoma {eco:0000269 golgi apparatus using this antibody. Is that expected? Could you tell me where is POSTN supposed to be expressed?

According to literature, osteosarcoma eco:0000269 does express POSTN. Generally POSTN expresses in golgi apparatus. Regarding which tissues have POSTN expression, here are a few articles citing expression in various tissues:

The anti-Periostin/Postn antibody (A01378) has not been validated for cross reactivity specifically with zebrafish tissues, though there is a good chance of cross reactivity.
