Background of Oncostatin M/OSM
OSM (ONCOSTATIN M) is a member of a cytokine family that includes leukemia-inhibitory factor, granulocyte colony-stimulating factor, and interleukin 6. This gene encodes a growth regulator which inhibits the proliferation of a number of tumor cell lines. It regulates cytokine production, including IL-6, G-CSF and GM-CSF from endothelial cells. OSM is mapped on 22q12.2. OSM has the ability to inhibit the growth of human A375 melanoma cells but not normal human fibroblasts. Treatment with recombinant OSM leads to the inhibition of proliferation and changes in cellular morphology of a number of tumor cell lines derived from a wide variety of tissue types. OSM also has the ability to inhibit the proliferation of murine M1 myeloid leukemic cells and can induce their differentiation into macrophage-like cells, a function shared by LIF, CSF3, and IL6. The direction of gene transcription was telomeric to centromeric, with the OSM gene located upstream of the LIF gene.

Would PA1668 anti-Oncostatin M/OSM antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

You can see on the product datasheet, PA1668 anti-Oncostatin M/OSM antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Our lab technicians have not tested anti-Oncostatin M/OSM antibody PA1668 on monkey. You can run a BLAST between monkey and the immunogen sequence of anti-Oncostatin M/OSM antibody PA1668 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated monkey samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in monkey lung in WB, you can get your next antibody for free.
