Background of IL-17C
IL17C, also known as CX2, is a protein that in humans is encoded by the IL17C gene. IL17C is mapped to 16q24.3. The protein encoded by this gene is a T cell-derived cytokine that shares the sequence similarity with IL17. This cytokine was reported to stimulate the release of tumor necrosis factor alpha and interleukin 1 beta from a monocytic cell line. The expression of this cytokine was found to be restricted to activated T cells. IL17C is an essential autocrine cytokine regulating innate epithelial immune responses. It also plays an important role in the pathogenesis of inflammatory arthritis.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: mouse spleen tissue lysates,
Lane 2: mouse thymus tissue lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-IL17C antigen affinity purified polyclonal antibody (Catalog # A00164-2) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for IL17C at approximately 29KD. The expected band size for IL17C is at 22KD.

Our lab technicians have not tested anti-IL17C antibody A00164-2 on horse. You can run a BLAST between horse and the immunogen sequence of anti-IL17C antibody A00164-2 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated horse samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in horse quadriceps femoris in ELISA, you can get your next antibody for free.
