Background of IL-11
Interleukin 11 (IL-11) is a protein that in humans is encoded by the IL11 gene. The protein encoded by this gene is a member of the gp130 family of cytokines. This gene is mapped to 19q13.42. It is a key regulator of multiple events in hematopoiesis, most notably the stimulation of megakaryocyte maturation. IL-11 has been demonstrated to improve platelet recovery after chemotherapy-induced thrombocytopenia, induce acute phase proteins, modulate antigen-antibody responses, participate in the regulation of bone cell proliferation and differentiation and could be use as a therapeutic for osteoporosis. What's more, IL-11 can stimulate the growth of certain lymphocytes and it has functions in many other tissues, including the brain, gut, testis and bone. In transgenic mice, overexpression of the human IL11 gene resulted in the stimulation of bone formation to increase cortical thickness and strength of long bones.
Antibody Validation

Figure 1. Western blot analysis of IL11 using anti-IL11 antibody (A01160-1).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: rat liver tissue lysates,
Lane 2: rat kidney tissue lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-IL11 antigen affinity purified polyclonal antibody (Catalog # A01160-1) at 0.5 ug/mL overnight at 4 then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for IL11 at approximately 21KD. The expected band size for IL11 is at 21KD.

Asked: 2020-01-13
Answer
From literature bone marrow does express IL11. From Uniprot.org, IL11 is expressed in adenohypophysis, bone marrow, prostate, among other tissues. Regarding which tissues have IL11 expression, here are a few articles citing expression in various tissues:
Bone marrow, Pubmed ID: 1828438
Prostate, Pubmed ID: 15489334

Question
Our team were satisfied with the WB result of your anti-IL11 antibody. However we have seen positive staining in adenohypophysis secreted using this antibody. Is that expected? Could you tell me where is IL11 supposed to be expressed?

Question
We are currently using anti-IL11 antibody A01160-1 for rat tissue, and we are content with the ELISA results. The species of reactivity given in the datasheet says mouse, rat. Is it possible that the antibody can work on monkey tissues as well?
J. Anderson
Verified customer
Asked: 2018-07-24
Answer
The anti-IL11 antibody (A01160-1) has not been tested for cross reactivity specifically with monkey tissues, but there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in monkey you can get your next antibody for free. Please contact me if I can help you with anything.
