Background of Cd2ap

CD2-associated protein is a protein that in humans is encoded by the CD2AP gene. This gene encodes a scaffolding molecule that regulates the actin cytoskeleton. The protein directly interacts with filamentous actin and a variety of cell membrane proteins through multiple actin binding sites, SH3 domains, and a proline-rich region containing binding sites for SH3 domains. The cytoplasmic protein localizes to membrane ruffles, lipid rafts, and the leading edges of cells. It is implicated in dynamic actin remodeling and membrane trafficking that occurs during receptor endocytosis and cytokinesis. Haploinsufficiency of this gene is implicated in susceptibility to glomerular disease.

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Question

Does anti-Human CD2AP DyLight® 550 conjugated antibody A01756-Dyl550 work on bovine Flow Cytometry with platelet?

Verified Customer

Verified customer

Asked: 2019-09-24

Answer

Our lab technicians have not validated anti-Human CD2AP DyLight® 550 conjugated antibody A01756-Dyl550 on bovine. You can run a BLAST between bovine and the immunogen sequence of anti-Human CD2AP DyLight® 550 conjugated antibody A01756-Dyl550 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated bovine samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in bovine platelet in Flow Cytometry, you can get your next antibody for free.

Question

Would A01756-Dyl550 anti-Human CD2AP DyLight® 550 conjugated antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

D. Anderson

Verified customer

Asked: 2013-03-29

Answer

As indicated on the product datasheet, A01756-Dyl550 anti-Human CD2AP DyLight® 550 conjugated antibody as been validated on Flow Cytometry. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
