Background of EGFL6
EGFL6 is a member of the epidermal growth factor (EGF) repeat superfamily. Members of this superfamily are characterized by the presence of EGF-like repeats and are often involved in the regulation of cell cycle, proliferation, and developmental processes. The gene product contains a signal peptide, suggesting that it is secreted; an EGF repeat region consisting of 4 complete EGF-like repeats and 1 partial EGF-like repeat, 3 of which have a calcium-binding consensus sequence; an arg-gly-asp integrin association motif; and a MAM domain, which is believed to have an adhesive function. This gene is expressed early during development, and its expression has been detected in lung and meningioma tumors.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human U-87MG whole cell lysates,
Lane 2: human A431 whole cell lysates,
Lane 3: human HL-60 whole cell lysates,
Lane 4: human K562 whole cell lysates,
Lane 5: human THP-1 whole cell lysates,
Lane 6: human Hela whole cell lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-EGFL6 antigen affinity purified polyclonal antibody (Catalog # A08756) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for EGFL6 at approximately 62KD. The expected band size for EGFL6 is at 62KD.

Will anti-EGFL6 antibody A08756 work on bovine ELISA with brain?

Our lab technicians have not validated anti-EGFL6 antibody A08756 on bovine. You can run a BLAST between bovine and the immunogen sequence of anti-EGFL6 antibody A08756 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated bovine samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in bovine brain in ELISA, you can get your next antibody for free.

We are currently using anti-EGFL6 antibody A08756 for rat tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on monkey tissues as well?

The anti-EGFL6 antibody (A08756) has not been validated for cross reactivity specifically with monkey tissues, but there is a good chance of cross reactivity.
