Background of Siglec-3/CD33
CD33, also known as Siglec-3 (sialic acid binding Ig-like lectin 3, SIGLEC3, SIGLEC-3, gp67, p67), is a transmembrane receptor expressed on cells of myeloid lineage. It is usually considered myeloid-specific, but it can also be found on some lymphoid cells. CD33 binds sialic acids, therefore is a member of the SIGLEC family of lectins. By fluorescence in situ hybridization, CD33 is mapped to 19q13.3-q13.4.

CD33 was detected in paraffin-embedded sections of mouse spleen tissues using rabbit anti-CD33 Antigen Affinity purified polyclonal antibody (Catalog # RP1105) at 1 μg/mL. The immunohistochemical section was developed using SABC method (Catalog # SA1022).

Western blot analysis of CD33 expression in mosue lung extract (lane 1). CD33 at 45KD was detected using rabbit anti-CD33 Antigen Affinity purified polyclonal antibody (Catalog # RP1105) at 0.5 μg/mL. The blot was developed using chemiluminescence (ECL) method (Catalog # EK1002).

I took a look at the website and datasheets of our anti-CD33 antibody and it seems that RP1105 has been validated on mouse in both IHC and WB. Thus RP1105 should work for your application. From literature, leukocyte does express CD33. Generally CD33 expresses in isoform cd33m: cell membrane, isoform cd33m: peroxisome. Regarding which tissues have CD33 expression, here are a few articles citing expression in various tissues:

My question regarding product RP1105, anti-CD33 antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

We suggest not storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free RP1105 anti-CD33 antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.

It shows on the product datasheet, RP1105 anti-CD33 antibody as been tested on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
