Western blot analysis of different tissue lysates and recombinant protein solutions using chicken pAb to calbindin, M03047-2, dilution 1:5,000 in green: [1] protein standard (red), [2] rat cerebellum, [3] pig hippocampus, [4] cow cerebellum, [5] protein standard (red). Next lanes are full length recombinant human proteins, [6] Parvalbumin, [7] Calretinin, [8] Calbindin. Bands at 25kDa in tissue lysates and ~30 kDa in protein solutions correspond to calbindin, the recombinant form being slightly larger due to the presence of a His tag and other vector derived sequence. The M03047-2 antibody specifically recognizes calbindin protein and does not react with the closely related proteins parvalbumin and calretinin.

Immunofluorescent analysis of rat cerebellum section stained with chicken pAb to calbindin, M03047-2, dilution 1:2,000, in green, and costained with rabbit pAb to MeCP2, dilution 1:5,000, in red. Following transcardial perfusion with 4% paraformaldehyde, brain was post fixed for 24 hours, cut to 45M, and free-floating sections were stained with above antibodies. Calbindin, often used as a Purkinje cell marker, is prominently detected in dendrites and perikarya of these cells in the cerebellar molecular layer. The MeCP2 antibody selectively stains nuclei of neuronal cells.

Calbindin was detected in a mice cerebellum. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 2 μg/ml rabbit anti-calbindin antibody (M03047-2) at 1:250 overnight at 4°C. Peroxidase Conjugated Goat Anti-rabbit IgG was used as secondary antibody at 1:250 and incubated for 2h at room temperature. The tissue section was developed using fluorescence.

"I tried this antibody for Purkinje cell labeling in mice cerebellum using IHC-P. Antigen retrieval - 20min boiling in EDTA based buffer. Permeabilization - 0.3% triton for 15min. Blocking - 10% goat serum for 1hr. Primary antibody - 1:250 overnight at 4 Degree C. Secondary antibody - Fluorescence conjugated secondary antibody at 1:250 for 2hr at RT. I was amazed to see the beautiful dendrites, soma and axons. In my hand, this is the best labeling of Purkinje cells I have ever seen using IHC-P. Beautiful labeling of Purkinje cell soma, dendrites and axons. Best antibody I have used till now. Highly recommended for IHC-P."

As indicated on the product datasheet, M03047-2 anti-Calbindin antibody as been validated on IF. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Based on literature kidney lung does express CALB1. Based on Uniprot.org, CALB1 is expressed in caudate nucleus, brain, amygdala, cerebellum substantia nigra, kidney lung, among other tissues. Regarding which tissues have CALB1 expression, here are a few articles citing expression in various tissues:

I have a question about product M03047-2, anti-Calbindin antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

We suggest not storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free M03047-2 anti-Calbindin antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
