Background of GDNF
Glial cell line-derived neurotrophic factor (GDNF) is a glycosylated, disulfide-bonded homodimer that is a distantly related member of the transforming growth factor-beta superfamily.1 GDNF, is a potent neurotrophic factor that promotes the survival of dopaminergic neurones in cultures including embryonic neuronal cultures.2 GDNF, in addition to its potential role in the differentiation and survival of central nervous system neurons, has profound effects on kidney organogenesis and the development of the peripheral nervous system.3 GDNF may have utility in the treatment of Parkinson's disease, which is marked by progressive degeneration of midbrain dopaminergic neurons.

Recommended Sample Dilution Ratios
According to our internal validation assays using this ELISA kit, to detect GDNF, Dilution ratio of 1:1, concentration in serum and plasma is less than the lowest standard, 31.2 pg/ml..
Some articles we found to cite concentrations of GDNF in samples: 14521734 (Pubmed IDs).
Intra Assay Consistency & Inter Assay Consistency
We measured random samples of Mouse GDNF ELISA Kit PicoKine® within the same batch/lot to ensure the consistency of the kits' performances. ELISA intra assay consistency is measured using wells from the same plate/assay kit. ELISA inter assay consistency is measured using wells from different plates from the same batch production/lot.

"We used this kit and the BDNF kit, because we were looking at neurogenesis within the hippocampus. We used the ELISA to complement our immunohistochemistry. It was easy to optimize the kits and the instructions were extremely easy to follow. Additionally, the instructions do not include using samples from tissue homogenates, but we did and the plates came out great. This product was selected after speaking with a representative about it at a conference. I am extremely glad we decided to go with this kit. This is a great kit. The protocol was easy to follow and optimize. We will be using these kits again."

6 Customer Q&As for Mouse GDNF ELISA Kit PicoKine®
Question
Q: Are Boster Bio recombinant proteins and antibodies sterile?
Verified Customer
Verified customer
Asked: 2020-04-14
Answer
A: although the vials are bottled using aseptic techniques, heat-treated vials, and sterile stock solutions, they are not considered or guaranteed to be sterile. If sterile material is a must for an experiment, the material can be filtered through a 0.2 micron filter designed for use with biological fluids.

Question
Would EK0935 work on tissue homogenate samples that has been prepared with RIPA buffer?
Verified customer
Asked: 2019-04-01
Answer
Mouse GDNF ELISA Kit PicoKine™ (EK0935) hasn't been tested on tissue homogenates. Endogenous biotin in tissue homogenates might introduce false positive signal. We recommend to run a pilot test as described below to confirm the affect of endogeous biotin first. Add tissue homogenates into the wells and then add ABC abd TMB without adding any biotinylated detection antibody to see if any signal will be observed. If no signal is produced, proceed and work on the tissue sample by using the kit.

Question
Q: if the enzyme conjugated GDNF antibodies are mixed with the substrate, will that change the substrate into the enzymatic reaction product? Or the enzyme function is only activated when the antibody is attached with the GDNF antigen?
Verified Customer
Verified customer
Asked: 2018-05-21
Answer
A: The enzyme is always active. Avoid contaminating the substrate with enzyme prior to the incubation otherwise it compromises the assay with false positive signal.

Question
Q: how long can samples (cell cultures, serum, and plasma) be stored and still be stable for analysis GDNF using the EK0935 Mouse GDNF Picokine® ELISA Kit?
E. Liu
Verified customer
Asked: 2014-11-24
Answer
A: Boster Bio does not evaluate sample stability. Variations in sample collection, processing, and storage may change the stabilityof samples. It is recommend to assay sample immediately after collection when possible, or aliquot into single use volumes and store samples frozen. limit repetitive freeze-thaw cycles with the stored samples to limit protein degradation.
