Background of IL-10
Interleukin-10, also called cytokine synthesis inhibitory factor, is implicated in tumorigenesis, and it has been shown that polymorphisms in its gene promoter correlate with differential amounts of production. IL-10 is an important cytokine with anti-inflammatory, anti-immune, and antifibrotic functions. It is also an important regulatory cytokine whose involvement extends into diverse areas of the human immune system. IL-10 is a recently described natural endogenous immunosuppressive cytokine that has been identified in human, murine, and other organisms. IL-10 significantly affects chemokine biology, because human IL-10 inhibits chemokine production and is a specific chemotactic factor for CD8+ T cells. It suppresses the ability of CD4+ T cells, but not CD8+ T cells, to migrate in response to IL-8. Interleukin-10 gene polymorphisms and interleukin-10 production capability may contribute to the development of skin squamous cell carcinomas after renal transplantation.1 The interleukin-10 locus contributes to the heritability of psoriasis susceptibility. With regard to sudden infant death, IL-10 is of special interest. This is an immunoregulatory cytokine that plays an important role in the development of infectious disease The mIL-10 gene is mapped to mouse chromosome 1 and the hIL-10 gene is also mapped to human chromosome 1. The standard product used in this kit is recombinant human IL-10, consisting of 160 amino acids with the molecular mass of 18.6KDa.

Q: if the enzyme conjugated IL-10 antibodies are mixed with the substrate, will that change the substrate into the enzymatic reaction product? Or the enzyme function is only activated when the antibody is attached with the IL-10 antigen?

A: The enzyme is always active. Avoid contaminating the substrate with enzyme prior to the incubation otherwise it compromises the assay with false positive signal.

Q: can you recommend the dilution ratio of serum samples for detection of IL-10 in Human plasma? I am trying to measure a a number of analytes and it requires 100ul of diluted samples for each well. We have low serum quantitys so we like to dilute as much as possible.

A: unable to understand the physiological or pathological context of your samples we cannot recommend a dilution ratio without performing a pilot test with your samples. Here is how you can perform a pilot study on your own: perform a serial dilution of your samples on the IL-10 ELISA kit to make sure you have a linear ascending curve followed by a plateau, which signifies the samples saturating the detection limit of the kit. Then you can pick the dilution ratios from samples in the linear part of the curve as your experimental dilution ratio.

A: Unfortunately, Boster Bio has not routinely validated tissue homogenates as a sample type for ELISA kits. This does not mean that ELISA kits are not suitable for other sample types than we have tested: it means further investigation is required. One will need to perform a spike and recovery study to determine if an unvalidated sample type will work with a particular kit. To perform a spike and recovery experiment, one should divide a sample into two aliquots. In one of the aliquots, the user should spike in a known amount of the kit standard. a dilution series is performed comparing the spiked versus the unspiked sample. Generally, samples with expected recovery and linearity between 80-120% are considered acceptable. This method can be used to validate any sample type that has not been evaluated by Boster Bio. for a more detailed spike and recovery protocol, please contact technical support.
Note: acceptable ranges should be determined individually by each laboratory. Additionally, technical support can help determine if a buffer component is not compatible with a given ELISA kit. please view the Citations tab on the product webpage for peer-reviewed papers utilizing a wide range of sample types. The Human IL-6 EK0410 Elisa Kit and Human IL-10 EK0416 Elisa Kit have not been tested on tissue homogenates. Endogenous biotin in tissue homogenates might introduce a false positive signal. We recommend to run a pilot test to confirm the effect of the endogenous biotin first. Add tissue homogenates into the wells and then add ABC and TMB without adding any biotinylated detection antibody to see if any signal will be observed. If no signal is produced, proceed and work on the tissue sample by using the kit.

A: Chelating agents such as EDTA, Heparin and Citrate can sequester metal ions from the functional domain of IL-10 causing degradation of its protein structure. IL-10 may be denatured as a result and may compromise the assay's measurements. The chilating sites could also be too close to the epitopes a must for detection and block the antigen antibody reaction. We have tested the IL-10 ELISA, treating samples with a number of anticoagulants and decided that heparin, EDTA or citrate can be used for treatment of blood/plasma samples. Do not use other anticoagulents when collecting samples.

For untreated cell supernatants, the suggested dilution ratio is 1:1. For treated cell supernatants, concentrations of the samples can vary greatly. We suggest the customer run pilot tests for different group samples to figure out the optimal dilution ratio.

Components in lysate and lysis buffer can have an affect on immunoreactivity, so if lysate is not a validated sample type, care must be taken in sample preparation and validation.

A: we do not recommend freezing and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test IL-10 for a later time, you should let the blood clot in glass tubes and collect the serum to freeze for later use.
