Background of UBE2N/Ubc13
Ubiquitin-conjugating enzyme E2 N is a protein that in humans is encoded by the UBE2N gene. The modification of proteins with ubiquitin is an important cellular mechanism for targeting abnormal or short-lived proteins for degradation. Ubiquitination involves at least three classes of enzymes: ubiquitin-activating enzymes, or E1s, ubiquitin-conjugating enzymes, or E2s, and ubiquitin-protein ligases, or E3s. This gene encodes a member of the E2 ubiquitin-conjugating enzyme family. Studies in mouse suggest that this protein plays a role in DNA postreplication repair.
Antibody Validation
Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Lane 3: human RT4 whole cell lysates,
Lane 4: human HEL whole cell lysates,
Lane 5: rat heart tissue lysates,
Lane 6: rat thymus tissue lysates,
Lane 7: rat C6 whole cell lysates,
Lane 8: mouse thymus tissue lysates,
Lane 9: mouse small intestine tissue lysates,
Lane 10: mouse NIH/3T3 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Ubc13/UBE2N antigen affinity purified polyclonal antibody (Catalog # A02462-2) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Ubc13/UBE2N at approximately 17 kDa. The expected band size for Ubc13/UBE2N is at 17 kDa.
