Background of TREX1
Three prime repair exonuclease 1 is an enzyme that in humans is encoded by the TREX1 gene. This gene encodes a nuclear protein with 3' exonuclease activity. The encoded protein may play a role in DNA repair and serve as a proofreading function for DNA polymerase. It is also a component of the SET complex, and acts to rapidly degrade 3' ends of nicked DNA during granzyme A-mediated cell death. Mutations in this gene result in Aicardi-Goutieres syndrome, chilblain lupus, Cree encephalitis, and other diseases of the immune system. Alternative splicing results in multiple transcript variants.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 40 ug of sample under reducing conditions.
Lane 1: SMMC Whole Cell Lysate.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-TREX1 antigen affinity purified polyclonal antibody (Catalog # PB9748) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for TREX1 at approximately 33 kDa. The expected band size for TREX1 is at 39 kDa.

We are currently using anti-TREX1 antibody PB9748 for human tissue, and we are well pleased with the WB results. The species of reactivity given in the datasheet says human. Is it true that the antibody can work on monkey tissues as well?

The anti-TREX1 antibody (PB9748) has not been tested for cross reactivity specifically with monkey tissues, but there is a good chance of cross reactivity.
