Background of TRAP1

The 90 kDa heat shock protein (HSP90) family of proteins that play an important physiological role. HSP90 is involved in numerous cellular processes but is best known for its association with signal transduction machinery. A recently cloned homolog of HSP90 is the tumor necrosis factor receptor-associated protein (TRAP1). Like HSP90, TRAP1 is found to be associated with numerous proteins involved in diverse actions (1, 2). Immunofluorescence data has shown TRAP1 to be localized in the mitochondria of mammalian cells. This observation and the fact that TRAP1 is shown to have a mitochondrial targeting pre-sequence strongly implicates TRAP1 as a mitochondrial matrix protein (3).

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-TRAP1 antigen affinity purified polyclonal antibody (Catalog # M02426-3) at 0.5 ug/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-Mouse IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # SA1021) with Tanon 5200 system. A specific band was detected for TRAP1.

Fixation: 4% Formaldehyde for 15 min at RT. Primary Antibody: Mouse Anti-Trap1 Monoclonal Antibody (M02426-3) at 1:100 for 60 min at RT. Secondary Antibody: Goat Anti-Mouse ATTO 488 at 1:100 for 60 min at RT. Counterstain: DAPI (blue) nuclear stain at 1:5000 for 5 min RT. Localization: Nucleus. Magnification: 60X.
