Background of Tau

Tau is a key microtubule-associated protein that plays an important role in the formation of microtubules in axons (Binder et al. 1985). Six tau isoforms have been identified as products of a single gene produced by alternative mRNA splicing (Goedert 1990). Tau mutations have been implicated in many neurodegenerative disorders such as Alzheimer’s disease (AD), Pick’s disease and progressive supranuclear palsy. It has been well documented that hyperphosphorylated tau is a major component of paired helical filaments in AD brain (Lee 1995). Ser-416 has been demonstrated to be a major phosphorylation site in vitro by CaM kinase II (Steiner at al. 1990).

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

3 Customer Q&As for Anti-Phospho-Tau (Ser416) MAPT Antibody

Question

We were well pleased with the WB result of your anti-Phospho-Tau (Ser416) antibody. However we have been able to see positive staining in fetal brain cytosol using this antibody. Is that expected? Could you tell me where is MAPT supposed to be expressed?

Question

We are currently using anti-Phospho-Tau (Ser416) antibody P00097 for mouse tissue, and we are content with the WB results. The species of reactivity given in the datasheet says mouse, rat. Is it likely that the antibody can work on primate tissues as well?

M. Collins

Asked: 2014-03-10

Answer

The anti-Phospho-Tau (Ser416) antibody (P00097) has not been tested for cross reactivity specifically with primate tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in primate you can get your next antibody for free. Please contact me if I can help you with anything.
