Background of Synaptotagmin 1
Synaptotagmin is widely regarded as the primary calcium sensor for synaptic vesicle exocytosis (Fernandez-Chacon et al., 2001; Wang et al., 2003). Moreover, recent studies indicate that the protein also plays a key role in endocytosis (Poskanzer et al., 2003). Synaptotagmin can be phosphorylated by multiple protein kinases and this may play a key role in modulation of synaptotagmin’s ability to influence both the exocytotic and endocytotic components of synaptic transmission (Hilfiker et al., 1999; Lee et al., 2004).
Antibody Validation
Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Western blot of rat cortical lysate showing specific immunolabeling of the ~62 kDa synaptotagmin phosphorylated at Thr202 in the first lane (-). Phosphospecificity is shown in the second lane (+) where the immunolabeling is completely eliminated by blot treatment with lambda phosphatase (gamma-Ptase, 1200 units for 30 minutes).

4 Customer Q&As for Anti-Phospho-Synaptotagmin (Thr202) Syt1 Antibody
Question
We are currently using anti-Phospho-Synaptotagmin (Thr202) antibody P02314 for rat tissue, and we are satisfied with the IHC results. The species of reactivity given in the datasheet says rat. Is it possible that the antibody can work on primate tissues as well?

Answer
The anti-Phospho-Synaptotagmin (Thr202) antibody (P02314) has not been tested for cross reactivity specifically with primate tissues, but there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in primate you can get your next antibody for free. Please contact me if I can help you with anything.

Answer
From literature amygdala brain does express SYT1. From Uniprot.org, SYT1 is expressed in middle temporal gyrus, amygdala brain, hippocampus, pns, among other tissues. Regarding which tissues have SYT1 expression, here are a few articles citing expression in various tissues:
Amygdala, and Brain, Pubmed ID: 14702039
Hippocampus, Pubmed ID: 17974005
PNS, Pubmed ID: 15489334

Question
We ordered your anti-Phospho-Synaptotagmin (Thr202) antibody for WB on hippocampus last year. I am using rat, and We are going to use the antibody for IHC next. My question regards examining hippocampus as well as amygdala brain in our next experiment. Do you have any suggestion on which antibody would work the best for IHC?

Answer
I have checked the website and datasheets of our anti-Phospho-Synaptotagmin (Thr202) antibody and it seems that P02314 has been tested on rat in both WB and IHC. Thus P02314 should work for your application. Our Boster satisfaction guarantee will cover this product for IHC in rat even if the specific tissue type has not been validated. We do have a comprehensive range of products for IHC detection and you can check out our website bosterbio.com to find out more information about them.

Question
Our lab were well pleased with the WB result of your anti-Phospho-Synaptotagmin (Thr202) antibody. However we have been able to see positive staining in amygdala brain cytoplasmic vesicle using this antibody. Is that expected? Could you tell me where is SYT1 supposed to be expressed?
