Background of Synaptotagmin 1
The alpha-crystallins are major water-soluble lens structural proteins of the vertebrate eye that are related to the small heat shock protein family. The alpha-crystallins possess structural and functional similarities with HSP25 and HSP27 (1). Mammalian lens cystallins are divided into alpha, beta and gamma families. Alpha and beta families are further divided into acidic and basic groups (Alpha-A and Alpha-B respectively). In the lens, alpha-crystallin primarily functions to maintain proper refractive index, however it can also function as a molecular chaperone that binds to the denatured proteins, keeping them in solution and thereby maintaining the translucency of the lens. When cellular stress occurs, alpha-crystallin enters its' phosphorylated state and may serve a structural control function and play a role in protein maintenance (2). In addition to their interaction with proteins, alpha-crystallins also interact with native molecules such as membrane proteins, Golgi matrix protein, structural proteins, nuclear proteins and DNA (3, 4, 5, 6, and 7). Two other functions are an autokinase activity and participation in the intracellular architecture, and it has also been proven that both alpha-A and B prevent apoptosis by inhibiting caspases (8).

Western blot analysis of Synaptotagmin expression in MW marker (lane 1), mouse brain extract (lane 2) and rat brain extract (lane 3). Synaptotagmin at 65KD was detected using mouse anti-Synaptotagmin Antigen Affinity purified monoclonal antibody (Catalog # M02314) at 1: 1000. The blot was developed using colorimetric method.

Based on literature, pns does express SYT1. Generally SYT1 expresses in cytoplasmic vesicle, secretory vesicle. Regarding which tissues have SYT1 expression, here are a few articles citing expression in various tissues:

As indicated on the product datasheet, M02314 anti-Synaptotagmin antibody (Monoclonal, ASV30) as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Can you help my question with product M02314, anti-Synaptotagmin antibody (Monoclonal, ASV30). I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

It is not recommended storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free M02314 anti-Synaptotagmin antibody (Monoclonal, ASV30), we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
