Background of SDHA
SDHA (Succinate dehydrogenase complex, subunit a, flavoprotein), also called SDH1, HOMOLOG OF, is a protein that in humans is encoded by the SDHA gene. The SDHA gene is mapped on 5p15.33. The SDHA gene is highly polymorphic. The gene that codes for the SDHA protein is nuclear, even though the protein is located in the inner membrane of the mitochondria. Heterozygous carriers of an SDHA mutation do not developparagangliomas as has been seen for mutations in the other subunits. In vitro functional expression studies in the yeast homolog showed that the mutation resulted in a loss of SDH activity and rendered the mutant SDHA protein more susceptible to proteolysis. The findings indicated that SDHA, like other SDH subunits, can act as a tumor suppressor gene.
Antibody Validation
Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Figure 1. Western blot analysis of SDHA using anti-SDHA antibody (PA1717).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human 293T whole cell lysates,
Lane 2: human MCF-7 whole cell lysates,
Lane 3: human HepG2 whole cell lysates,
Lane 4: rat brain tissue lysates,
Lane 5: mouse brain tissue lysates,
Lane 6: mouse spleen tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-SDHA antigen affinity purified polyclonal antibody (Catalog # PA1717) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for SDHA at approximately 73 kDa. The expected band size for SDHA is at 73 kDa.

Figure 2. IHC analysis of SDHA using anti-SDHA antibody (PA1717).
SDHA was detected in a paraffin-embedded section of human lung cancer tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 2 μg/ml rabbit anti-SDHA Antibody (PA1717) overnight at 4°C. Peroxidase Conjugated Goat Anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. Figure 7. Flow Cytometry analysis of PC-3 cells using anti-SDHA antibody (PA1717).
Overlay histogram showing PC-3 cells stained with PA1717 (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer.
