Figure:Lysates of normal lung fibroblasts (WI38) and of human fibrosarcoma (8387) were probed with the PTX3, pAb, (Prod. No. A00649), over night at +4 °C at a concentration of 500ng/ml in 5% dry milk in PBS+/+ containing0.05% Tween 20. The signal was revealed with an HRP-conjugated anti-rat IgG secondary antibody in 5% dry milk PBS+/+ containing 0.05% Tween 20 after incubation for 1 hour at room temperature and IMMOBILON Western Chemiluminescent substrate from Millipore.

PTX3 was detected in paraffin-embedded section. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1ug/ml rabbit anti-PTX3 Antibody (A00649) overnight at 4°C. Biotinylated goat anti Rabbit IgG antibody was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC)(Catalog # SA1022) with DAB as the chromogen.
