Background of PKC epsilon
Protein kinase C epsilon type, also known as PKCE, is an enzyme that in humans is encoded by the PRKCE gene. The protein encoded by this gene is one of the PKC family members. PRKCE is mapped to 2p21. This kinase has been shown to be involved in many different cellular functions, such as apoptosis, cardioprotection from ischemia, heat shock response, as well as insulin exocytosis. It has been found that activation of PRKCE can induce VR1 channel activity at room temperature in the absence of any other agonist. PRKCE gene plays a role in apoptosis signaling pathways in thyroid cells and it has been indicated that a naturally occurring PRKCE mutant that functions as a dominant negative can block cell death triggered by a variety of stimuli. Expression of PRKCE inhibits chemotherapy-induced caspase-3 activation and apoptosis, thereby leading to cell survival.
Antibody Validation
Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Figure 1. Western blot analysis of PRKCE using anti-PRKCE antibody (A01151-1).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: rat brain tissue lysates,
Lane 2: mouse lung tissue lysates,
Lane 3: mouse brain tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PRKCE antigen affinity purified polyclonal antibody (Catalog # A01151-1) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for PRKCE at approximately 94 kDa. The expected band size for PRKCE is at 83 kDa.

Figure 2. IF analysis of PRKCE4 using anti-PRKCE4 antibody (A01151-1) and anti-Tubulin Alpha antibody (M03989-3).
PRKCE4 was detected in immunocytochemical section of A549 cell. Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent (AR0022) for 15 mins. The cells were blocked with 10% goat serum. And then incubated with 5 μg/mL rabbit anti-PRKCE4 Antibody (A01151-1) and mouse anti-Tubulin Alpha antibody (M03989-3) overnight at 4°C. DyLight®488 Conjugated Goat Anti-Rabbit IgG (BA1127) and Cy3 Conjugated Goat Anti-Mouse IgG (BA1031) were used as secondary antibody at 1:500 dilution and incubated for 30 minutes at 37°C. Visualize using a fluorescence microscope and filter sets appropriate for the label used.
