Background of Proprotein Convertase 9/PCSK9
Proprotein convertase subtilisin/kexin type 9, also known as PCSK9, is an enzyme that in humans is encoded by the PCSK9 gene. This gene encodes a proprotein convertase belonging to the proteinase K subfamily of the secretory subtilase family. By genomic sequence analysis, PCSK9 was mapped to chromosome 1p32. This gene is a crucial player in the regulation of plasma cholesterol homeostasis. It may prevent the recycling of LDLR from endosomes to the cell surface or direct it to lysosomes for degradation. PCSK9 can induce ubiquitination of LDLR leading to its subsequent degradation. This gene is involved in the disposal of non-acetylated intermediates of BACE1 in the early secretory pathway.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human A431 whole cell lysates,
Lane 2: human HepG2 whole cell lysates,
Lane 3: human MCF-7 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PCSK9 antigen affinity purified polyclonal antibody (Catalog # PA2130) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for PCSK9 at approximately 66 kDa. The expected band size for PCSK9 is at 74 kDa.

PCSK9 was detected in a paraffin-embedded section of human liver cancer tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 2 μg/ml rabbit anti-PCSK9 Antibody (PA2130) overnight at 4°C. Peroxidase Conjugated Goat Anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using HRP Conjugated Rabbit IgG Super Vision Assay Kit (Catalog # SV0002) with DAB as the chromogen.

From what I have seen in literature cervix carcinoma erythroleukemia does express PCSK9. From what I have seen in Uniprot.org, PCSK9 is expressed in right lobe of liver, cerebellum teratocarcinoma, cervix carcinoma, cervix carcinoma erythroleukemia, among other tissues. Regarding which tissues have PCSK9 expression, here are a few articles citing expression in various tissues:

Our lab technicians have not validated anti-PCSK9 antibody PA2130 on dog. You can run a BLAST between dog and the immunogen sequence of anti-PCSK9 antibody PA2130 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated dog samples, this use of the antibody is not covered by our guarantee. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

We suggest not storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free PA2130 anti-PCSK9 antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.

You can see on the product datasheet, PA2130 anti-PCSK9 antibody as been tested on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
