Background of PAR2
Protease activated receptor 2 (PAR2), also known as coagulation factor II (thrombin) receptor-like 1 (F2RL1) or G-protein coupled receptor 11 (GPR11), is a protein that in humans is encoded by the F2RL1 gene. F2RL1 is a member of the large family of 7-transmembrane-region receptors that couple to guanosine-nucleotide-binding proteins. F2RL1 is also a member of the protease-activated receptor family. It is activated by trypsin, but not by thrombin. It is activated by proteolytic cleavage of its extracellular amino terminus. The new amino terminus functions as a tethered ligand and activates the receptor. The F2RL1 gene contains two exons and is widely expressed in human tissues. Additionally, PAR2 modulates inflammatory responses and acts as a sensor for proteolytic enzymes generated during infection.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 40ug of sample under reducing conditions.
Lane 1: HELA Whole Cell Lysate,
Lane 2: COLO320 Whole Cell Lysate,
Lane 3: SW620 Whole Cell Lysate,
Lane 4: HEPG2 Whole Cell Lysate.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PAR2 antigen affinity purified polyclonal antibody (Catalog # PB9700) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for PAR2 at approximately 49 kDa. The expected band size for PAR2 is at 44 kDa.

As indicated on the product datasheet, PB9700 anti-PAR2/F2RL1 antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
