Storage & Handling

Background of IGFBP-5

Insulin-like growth factor-binding protein 5 is a protein that in humans is encoded by the IGFBP5 gene. The expression of IGFBP5 by stable transfection and adenovirus-mediated infection is inhibitory to growth in 2 human breast cancer cell lines. IGFBP5 expression leads to G2/M cell cycle arrest and apoptosis. Stable expression of IGFBP5 in the breast cancer cell lines also inhibits the formation and growth of tumors following injection in athymic mice. It is concluded that IGFBP5 is a growth inhibitor and proapoptotic agent in breast cancer cells. Additionally, IGFBP-5 is expressed by fibroblasts, myoblasts and osteoblasts, making it the predominant IGFBP found in bone extracts. It has a strong affinity for hydroxyapatite, allowing it to bind to bone cells. When bound to extracellular matrix, IGFBP-5 is protected from proteolysis and potentiates IGF activity, but when it is soluble, IGFBP-5 is cleaved to a biologically inactive 21 kDa fragment (1, 2).

Does PB9711 anti-IGFBP5 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Answer

It shows on the product datasheet, PB9711 anti-IGFBP5 antibody as been tested on ELISA. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

We are currently using anti-IGFBP5 antibody PB9711 for human tissue, and we are satisfied with the ELISA results. The species of reactivity given in the datasheet says human. Is it likely that the antibody can work on pig tissues as well?

Our lab technicians have not tested anti-IGFBP5 antibody PB9711 on primate. You can run a BLAST between primate and the immunogen sequence of anti-IGFBP5 antibody PB9711 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated primate samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in primate placenta in ELISA, you can get your next antibody for free.
