Background of Blood group H inhibitor
Galactoside 2-alpha-L-fucosyltransferase 1 is an enzyme that in humans is encoded by the FUT1 gene. It is mapped to 19q13.3. The protein encoded by this gene is a Golgi stack membrane protein that is involved in the creation of a precursor of the H antigen, which is required for the final step in the soluble A and B antigen synthesis pathway. This gene is one of two encoding the galactoside 2-L-fucosyltransferase enzyme. Mutations in this gene are a cause of the H-Bombay blood group.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Rat Kidney Tissue Lysate at 50ug,
Lane 2: SW620 Whole Cell Lysate at 40ug,
Lane 3: A549 Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-FUT1 antigen affinity purified polyclonal antibody (Catalog # PB9593) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for FUT1 at approximately 50 kDa. The expected band size for FUT1 is at 50 kDa.

FUT1 was detected in a paraffin-embedded section of mouse intestine tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1 μg/ml rabbit anti-FUT1 Antibody (PB9593) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) (Catalog # SA1022) with DAB as the chromogen.

We are currently using anti-Galactoside 2-alpha-L-fucosyltransferase 1/FUT1 antibody PB9593 for human tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it true that the antibody can work on horse tissues as well?

The anti-Galactoside 2-alpha-L-fucosyltransferase 1/FUT1 antibody (PB9593) has not been tested for cross reactivity specifically with horse tissues, though there is a good chance of cross reactivity.
