Background of EED
Polycomb protein EED is a protein that in humans is encoded by the EED gene. It is a member of the Polycomb-group (PcG) family. PcG family members form multimeric protein complexes, which are involved in maintaining the transcriptional repressive state of genes over successive cell generations. This protein interacts with enhancer of zeste 2, the cytoplasmic tail of integrin β7, immunodeficiency virus type 1 (HIV-1) MA protein, and histone deacetylase proteins. Furthermore, this protein mediates repression of gene activity through histone deacetylation, and may act as a specific regulator of integrin function. Two transcript variants encoding distinct isoforms have been identified for this gene.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Rat Cardiac Muscle Tissue Lysate at 50ug,
Lane 2: Rat Kidney Tissue Lysate at 50ug,
Lane 3: Rat Liver Tissue Lysate at 50ug,
Lane 4: Rat Lung Tissue Lysate at 50ug,
Lane 5: SKOV Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-EED antigen affinity purified polyclonal antibody (Catalog # PB9581) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for EED at approximately 72 kDa. The expected band size for EED is at 51 kDa.

Question
We are currently using anti-EED antibody PB9581 for rat tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, rat. Is it possible that the antibody can work on canine tissues as well?

The anti-EED antibody (PB9581) has not been validated for cross reactivity specifically with canine tissues, though there is a good chance of cross reactivity.
