Background of CDC5L
Cell division cycle 5-like protein, also called PCDC5RP or CDC5-like, is a protein that in humans is encoded by the CDC5L gene. The CDC5L gene was mapped to chromosome 6p21 by fluorescence in situ hybridization. This DNA-binding protein involved in cell cycle control. It may act as a transcription activator. The Component of the PRP19-CDC5L complex forms an integral part of the spliceosome and is required for activating pre-mRNA splicing. The role of CDC5 (Polo-like kinase) in regulating Rho1 is likely to be relevant to cytokinesis and asymmetric cell division in other organisms.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human HEK293 whole cell lysates
Lane 2: human THP-1 whole cell lysates
Lane 3: rat C6 whole cell lysates
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CDC5L antigen affinity purified polyclonal antibody (Catalog # PA2123) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for CDC5L at approximately 100KD. The expected band size for CDC5L is at 92KD.

Question
We are currently using anti-CDC5L antibody PA2123 for rat tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, rat. Is it possible that the antibody can work on pig tissues as well?

The anti-CDC5L antibody (PA2123) has not been validated for cross reactivity specifically with pig tissues, but there is a good chance of cross reactivity.
