Background of DIS3
Exosome complex exonuclease RRP44 or Dis3 is an enzyme that in humans is encoded by the DIS3 gene. The DIS3 gene encodes a catalytic subunit of the exosome complex, an essential RNA degradation machinery involved in multiple aspects of RNA metabolism, including RNA processing, quality control, and turnover. DIS3 functions as an exonuclease, playing a key role in degrading various RNA species, such as messenger RNA (mRNA), ribosomal RNA (rRNA), and transfer RNA (tRNA). Its activity is crucial for maintaining cellular RNA homeostasis and gene expression regulation. Dysregulation of DIS3 expression or activity has been linked to various human diseases, including cancer and neurodegenerative disorders. Understanding the molecular mechanisms underlying DIS3 function is essential for elucidating its role in RNA metabolism and its potential implications for disease pathology and therapeutic targeting.
Antibody Validation
Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Figure 1. Western blot analysis of DIS3 using anti-DIS3 antibody (A01736-2).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human Hela whole cell lysates,
Lane 2: human Raji whole cell lysates,
Lane 3: human MCF-7 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-DIS3 antigen affinity purified polyclonal antibody (Catalog # A01736-2) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for DIS3 at approximately 109 kDa. The expected band size for DIS3 is at 109 kDa.
