Background of CD8
The CD8 antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell-cell interactions within the immune system. It is mapped to 2p11.2. The CD8 antigen, acting as a coreceptor, and the T-cell receptor on the T lymphocyte recognize antigen displayed by an antigen presenting cell (APC) in the context of class I MHC molecules. The functional coreceptor is either a homodimer composed of two alpha chains, or a heterodimer composed of one alpha and one beta chain. Both alpha and beta chains share significant homology to immunoglobulin variable light chains. This gene also encodes the CD8 alpha chain isoforms.

Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1μg/ml rabbit anti-CD8A Antibody (A02236-2) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC)(Catalog # SA1022) with DAB as the chromogen.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: rat thymus issue lysates,
Lane 2: rat thymus issue lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CD8A antigen affinity purified polyclonal antibody (Catalog # A02236-2) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for CD8A at approximately 26-35KD. The expected band size for CD8A is at 26KD.

From literature pericardium does express CD8A. From Uniprot.org, CD8A is expressed in thymus, pericardium, among other tissues. Regarding which tissues have CD8A expression, here are a few articles citing expression in various tissues:

My team were content with the WB result of your anti-CD8 alpha/Cd8a antibody. However we have seen positive staining in pericardium isoform 1: cell membrane using this antibody. Is that expected? Could you tell me where is CD8A supposed to be expressed?

We are currently using anti-CD8 alpha/Cd8a antibody A02236-2 for rat tissue, and we are well pleased with the WB results. The species of reactivity given in the datasheet says rat. Is it likely that the antibody can work on canine tissues as well?
