Background of CD5
CD5 is a member of the scavenger receptor cysteine-rich (SRCR) superfamily. Members of this family are secreted or membrane-anchored proteins mainly found in cells associated with the immune system. In humans, the gene is located on the long arm of chromosome 11. This protein is a type-I transmembrane glycoprotein found on the surface of thymocytes, T lymphocytes and a subset of B lymphocytes. The encoded protein contains three SRCR domains and may act as a receptor to regulate T-cell proliferation. Alternative splicing results in multiple transcript variants encoding different isoforms.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human Jurkat tissue lysates,
Lane 2: human CCRM-CEM whole cell lysates,
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with mouse anti-CD5 antigen affinity purified polyclonal antibody (Catalog # M00480-2) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-mouse IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1001) with Tanon 5200 system. A specific band was detected for CD5 at approximately 67KD. The expected band size for CD5 is at 54KD.

CD5 was detected in paraffin-embedded section of human tonsil cancer tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1μg/ml mouse anti-CD5 Antibody (M00480-2) overnight at 4°C. Biotinylated goat anti-mouse IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) (Catalog # SA1021) with DAB as the chromogen.

We are currently using anti-CD5 antibody (monoclonal, 4E2) M00480-2 for human tissue, and we are satisfied with the IHC-P results. The species of reactivity given in the datasheet says human. Is it possible that the antibody can work on horse tissues as well?

The anti-CD5 antibody (monoclonal, 4E2) (M00480-2) has not been tested for cross reactivity specifically with horse tissues, though there is a good chance of cross reactivity. Our lab were well pleased with the WB result of your anti-CD5 antibody (monoclonal, 4E2). However we have seen positive staining in pancreas cell membrane using this antibody. Is that expected? Could you tell me where is CD5 supposed to be expressed?

From literature, pancreas does express CD5. Generally CD5 expresses in cell membrane. Regarding which tissues have CD5 expression, here are a few articles citing expression in various tissues:

From what I have seen in literature thymus does express CD5. From what I have seen in Uniprot.org, CD5 is expressed in leukocyte, lymphocyte, thymus, pancreas, leukemic t-cell, among other tissues.
