Background of Nesprin-3
Component of SUN-protein-containing multivariate complexes also called LINC complexes which link the nucleoskeleton and cytoskeleton by providing versatile outer nuclear membrane attachment sites for cytoskeletal filaments. Involved in the maintenance of nuclear organization and structural integrity. Probable anchoring protein which tethers the nucleus to the cytoskeleton by binding PLEC which can associate with the intermediate filament system. Plays a role in the regulation of aortic epithelial cell morphology, and is required for flow-induced centrosome polarization and directional migration in aortic endothelial cells.

We are currently using anti-C14orf49 antibody A13164 for mouse tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, mouse. Is it true that the antibody can work on feline tissues as well?

The anti-C14orf49 antibody (A13164) has not been validated for cross reactivity specifically with feline tissues, though there is a good chance of cross reactivity. Will A13164 anti-C14orf49 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Answer
As indicated on the product datasheet, A13164 anti-C14orf49 antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
