Background of Lipocalin-2/NGAL

Lipocalin-2 (LCN2), also known as NGAL, is a protein associated with neutrophil gelatinase.1 The LCN2 gene is located at 9q34 and contains 7 exons.2 The 25-kD LCN2 protein is believed to bind small lipophilic substances such as bacteria-derived lipopolysaccharide (LPS) and formylpeptides and may function as a modulator of inflammation. NGAL tightly binds bacterial catecholate-type ferric siderophores through a cyclically permuted, hybrid electrostatic/cation-pi interaction and is a potent bacteriostatic agent in iron-limiting conditions.3 The primary LCN2 transcript is 3,696 nucleotides long, and the processed transcript is 809 nucleotides long.4 LCN2 expression in adult bone marrow, uterus, prostate, salivary gland, stomach, appendix, colon, trachea, and lung, and in fetal spleen and lung. The standard product used in this kit is recombinant rat NGAL, consisting of 177 amino acids with the molecular mass of 21kDa.

Our lab hasn't worked on stool supernatant samples with the Rat Lipocalin-2/NGAL ELISA Kit PicoKine™ (EK0855). We suggest to run a pilot test. In theory, the kit is able to detect the target protein if the expression level of NGAL in the stool supernatant samples falls in the assay range of EK0855.

A: do not freeze and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test Lipocalin-2 for a later time, you should let the blood clot in glass tubes and collect the serum to freeze for later use.

The recipe of our wash buffer is different from that of LB002-01. We haven't used the same buffer as LB002-01. And we do not suggest the customer use it. Please use the wash buffer included in the kit. Or prepare wash buffer as the method described below. Washing buffer (neutral PBS or TBS). ? Preparation of 0.01M TBS: Add 1.2g Tris, 8.5g Nacl; 450?l of purified acetic acid or 700?l of concentrated hydrochloric acid to 1000ml H2O and adjust pH to 7.2-7.6. Finally, adjust the total volume to 1L. ? Preparation of 0.01M PBS: Add 8.5g sodium chloride, 1.4g Na2HPO4 and 0.2g NaH2PO4 to 1000ml distilled water and adjust pH to 7.2-7.6. Finally, adjust the total volume to 1L.

Answer

A: Chelating agents such as EDTA, Heparin and Citrate can bind metal ions from the functional domain of Lipocalin-2 causing disruption of its protein structure. Lipocalin-2 may be denatured as a result and may compromise the assay's measurements. The chilating sites could also be too close to the epitopes a must for detection and block the antigen antibody reaction. We have tested the Lipocalin-2 ELISA, treating samples with different anticoagulants and decided that heparin can be used for treatment of blood/plasma samples. Do not use other anticoagulents when collecting samples.
